Combination of two powerful nutrients in wild grapes found to destroy cancer stem cells without harming healthy cells


Grapes might be one of the smaller fruits out there, but they punch far above their weight when it comes to providing health benefits. They’ve been revered for everything from their ability to make wine to their convenience and portability compared to other superfoods. Not content to stop there, however, this overachieving fruit is now making headlines for its potential to destroy cancer stem cells.
A team of researchers at Pennsylvania State University tested the effects of grape seed extract and a polyphenol compound found in the fruit known as resveratrol. Working on the theory that nearly all cancerous tumors are fueled by cancer stem cells, they examined 52 mice who had colon cancer tumors. The animals were divided into three groups for the study, one of which was fed the combination of grape compounds. A second group was given an anti-inflammatory drug that can reduce tumors known as sulindac, while the third group stuck to a normal diet and served as the control.

50 percent decrease in tumors

In the group of mice who were fed the grape compound, the number of tumors fell by an impressive 50 percent. While a similar decrease was seen in the group taking sulindac, that group suffered gastrointestinal toxicity; those taking the grape compound did not.
Interestingly, these two nutrients did not suppress the cancer stem cells as much when they were taken in small doses separately. Instead, researchers found that they got the best results when the two were combined. Best of all, the combination is not toxic to healthy cells, which means they could form the basis of a cancer treatment that is far safer than chemotherapy.
Resveratrol is found in the skin and seed of grapes, and you can also find it in foods like pomegranates, blueberries and cranberries. Past studies have shown that it can cut your risk of heart disease and keep some types of cancer at bay, like pancreatic cancer. Grape seed extract, meanwhile, has been found to help stave off tooth decay, kill leukemia cells, and enhance brain health.

Nature has the answers

This is just the latest example of nature providing the answer to one of life’s problems. Professor Vanamala, who led the study, said that plant-based diets provide the body with vital compounds that can kill cancer stem cells, so he recommends that people eat a variety of colorful vegetables and fruits to help prevent colon cancer, promote bacterial diversity, and prevent chronic illnesses. Of course, it’s important to seek out organic produce or you could be doing more harm than good.
Professor Vanamala pointed out that cultures that eat a plant-based diet typically have the lowest rates of colon cancer, and he feels this is because their food is covering all the bases by giving people a wide variety of compounds that can target the different pathways cancer stem cells depend on for their survival.
It is hoped that these compounds’ effects can be studied on humans and eventually lead to a new solution for protecting against colon cancer and preventing its recurrence in survivors. It couldn’t come at a better time as nearly 100,000 people are expected to develop the disease this year, according to estimates from the American Cancer Society. It’s the second most common type of cancer in women and the third most common in men. Finding a safe way to destroy these cells could prove to be a game-changer in the fight against cancer. Health, Wellness, and the Microwave Oven

The ubiquitous microwave oven, loved for its convenience in defrosting, reheating and preparing meals in a matter of minutes may not be quite the indispensable kitchen asset you once thought. Based on an alternating current, the microwave oven creates an electromagnetic radiation that forces the atoms, molecules and cells within the food to reverse polarity over a billion times per second. This remarkably high frequency creates an oscillating action of the molecules within the food and the high friction created as a result is the reason why microwave ovens can so quickly generate heat in food.

However, the cells and molecules within food are delicate and cannot withstand this violent force. As a result the food molecules are torn apart and rearranged into new chemical compositions that are foreign to the human body. Unable to recognize these new configurations or radiolytic products (unknown fusions, not found in nature and resulting from radiation) that are created as a result of the action of the microwaves, the body is not able to utilize it as food, and instead deposits it in and around the cells as toxic (or effectively nuclear) waste.

Years ago, when our son was in middle school, he had to complete a science project.  His project was to grow (from seed) and water two plants.  One seed received "standard" filtered tap water and the other seed received water that was first microwaved.  He photographed the results day by day.  The seed that received standard tap water grew as expected.  The seed that received microwaved water never sprouted.  After this project, we discarded our microwave oven.

Vitamin D protects against colon cancer: Study

A new study has confirmed the pivotal role of vitamin D, specifically its receptor (VDR), in slowing down the action of a key protein in the carcinogenic transformation process of colon cancer cells.

Researchers at the Vall d`Hebron Institute of Oncology (VHIO) collaborated with the Alberto Sols Institute of Biomedical Research (CSIC-UAB) for the study. 


This protein, known as beta-catenin, which is normally found in intestinal epithelial cells where it facilitates their cohesion, builds up in large quantities in other areas of the cells when the tumor transformation begins.
As a result of these changes, the protein is retained in the cell nucleus, where it facilitate the carcinogenic process, and this is the point at which vitamin D intervenes, or rather, the vitamin D receptor (VDR). 

“Our study has confirmed the pivotal role of the VDR in controlling the anomalous signal that sparks off the growth and uncontrolled proliferation of colon cells which, in the final instance, ends up causing a tumor to emerge,” said Hector Palmer, the coordinator of the study and head of the VHIO’s Stem Cells and Cancer laboratory. 

“The stimulation of this receptor suppresses the action of the beta-catenin protein, intercepting the series of events that change the intestinal cell into a malignant tumor cell,” he added. 

The researchers analyzed the effect of the VDR on human colon cancer cell cultures and observed that the concentration of the altered protein, beta-catenin, increased in cells without the VDR.
In light of these findings, chronic vitamin D deficiency represents a risk factor in the development of more aggressive colon tumors.

For Smokers, Low Levels of Vitamin D May Lead to Cancer

WASHINGTON, March 15, 2013 /PRNewswire/ -- New research appearing online today in Clinical Chemistry, the journal of AACC, shows that decreased levels of vitamin D may predispose smokers to developing tobacco-related cancer. This study illustrates that simple vitamin D blood tests and supplements have the potential to improve smokers' health.

In the U.S. alone, cigarette smoking accounts for more deaths annually than HIV, illegal drugs, alcohol, motor vehicle injuries, suicides, and murders combined. It is the primary causal factor for at least 30% of all cancer deaths, and can lead to multiple kinds of cancer, including bladder, cervical, esophageal, head and neck, kidney, liver, lung, pancreatic, and stomach, as well as myeloid leukemia. According to the Centers for Disease Control and Prevention, the healthcare expenditures and productivity losses due to smoking cost the economy approximately $193 billion per year. 

In this paper, Afzal et al. measured plasma vitamin D levels in blood samples collected in 1981–1983 from 10,000 Danes from the general population. Researchers then followed the study participants for up to 28 years through the Danish Cancer Registry. Of the participants, 1,081 eventually developed a tobacco-related cancer. The authors determined that the median vitamin D concentration among these participants was only 14.8 ng/mL, versus the higher 16.4 ng/mL median concentration found for all participants together.
These results show for the first time that the risk of tobacco-related cancers as a group is associated with lower concentrations of vitamin D. The data also indicate that tobacco smoke chemicals may influence vitamin D metabolism and function, while vitamin D may conversely modify the carcinogenicity of tobacco smoke chemicals. If further research confirms this, it would be consistent with previous studies demonstrating the anti-tumorigenic effects of vitamin D derivatives, as well as the correlation of vitamin D deficiency with favorable cancer-forming conditions and increased susceptibility to tobacco smoke carcinogens. Interestingly, though, low vitamin D levels were not connected with risk of other cancer types.
"Our analyses show that the association between lower concentrations of plasma vitamin D and higher risk of cancer may be driven by tobacco-related cancer as a group, which has not been shown before," stated author Borge G. Nordestgaard, MD, DMSc, in the paper. "This is important for future studies investigating the association between plasma vitamin D and risk of cancer."

Evidence Shows Vitamin D Helps Slow Cancer Cells

McGill University researchers have discovered vitamin D helps to slow the progression of cells of cancer from premalignant to malignant states.

The researchers reported in the journal Proceedings of the National Academy of Sciences vitamin D helps to inhibit both the production and function of the protein cMYC, which drives cell division and is active at elevated levels in more than half of all cancers.

Vitamin D has been known to have a wide range of physiological effects. Scientists have understood that a correlation exists between insufficient amounts of vitamin D and increased incidence of a number of cancers.
The correlations between the vitamin and cancers are strong for those cancers of the digestive tract, including colon cancer and certain forms of leukemia.

“For years, my lab has been dedicated to studying the molecular mechanisms of vitamin D in human cancer cells, particularly its role in stopping their proliferation,” Professor John White of the Faculty of Medicine’s Department of Physiology, said in a statement.

Vitamin D can be obtained from limited dietary sources as well as from direct exposure to the sun during the spring and summer months. Having a lack of exposure to the sun, mixed with a combination of poor dietary intake, has created vitamin D deficiency or insufficiency in large populations around the world. White said the team discovered vitamin D controls both the rate of production and the degradation of cMYC. They also found it strongly stimulates the production of a natural antagonist of cMYC known as MXD1, which shuts down cMYC function.

For the study, the researchers applied vitamin D to the skin of mice and observed a drop in the level of cMYC, discovering evidence of a decrease in its function. They also found strongly elevated levels of cMYC in a number of tissues, including skin and the lining of the colon.

“Taken together, our results show that vitamin D puts the brakes on cMYC function, suggesting that it may slow the progression of cells from premalignant to malignant states and keep their proliferation in check,” White said in the statement.

He said the team hopes the study will encourage people to maintain adequate vitamin D levels. Also, he said they hope the results will stimulate the development of large trials to test the effects of adequate supplementation.

A compound found in grapes can make prostate tumor cells more susceptible to radiation treatment, increasing a patient's chances of recovery, a University of Missouri researcher has found. 

Michael Nicholl, an assistant professor of surgical oncology in the MU School of Medicine, tested resveratrol on cells relatively resistant to radiation. He found that when the compound is added, the cells stopped growing and died at a much faster rate than when treated by either the compound or the radiation alone. 

Resveratrol is found in grape skins, red wine and also in over-the-counter pill form at health food stores.The finding isn't an excuse to up one's intake of wine, though. 

Although there might be some benefits to that, Nicholl stressed that his study only tested high doses delivered right to the tumor cells. In real-world scenarios, injecting large doses of resveratrol would cause harmful side effects. 

"What we really need to design is a nice delivery mechanism for higher doses to be effective," he said. Nicholl also discovered the resveratrol activated two proteins that further assisted in the fight against the cancer cells. 

The next step is to test the treatment in an animal model before clinical tests could be initiated. Nicholl anticipates studies on mice in the next several months. 

If additional studies are successful in the next few years, MU will request authority from the federal government to begin human drug testing that could lead to clinical trials. 

His initial study was published in the Journal of Andrology and Cancer Science.

Resveratrol promotes normal activity of SIRT-1. Studies have shown that SIRT-1 activates a critical component of calorie restriction in mammals and promotes fat mobilization. The most recent study (Nature 2006) demonstrated that obese mice fed red wine extract (resveratrol) were in better health and lived longer than obese mice that were not fed the red wine extract. The resveratrol group were more able to maintain heart health compared to mice that were not fed resveratrol. Resveratrol has also been shown to have phytoestrogen activity and may support optimal estradiol activity. Other studies have documented neuroprotective benefits associated with resveratrol supplementation.*

A comparison test was made to evaluate the activating effect on human SIRT-1 of Resveravine and pure trans-resveratrol. Only the trans-resveratrol form of the molecule promotes normal activation of the mammalian SIRT-1 gene in vitro test. Resveravine was more than eight times more likely to promote SIRT-1 activity than pure trans-resveratrol, showing that Resveravine is more efficient than resveratrol in promoting the normal activity of SIRT-1. Therefore, while the specific mechanisms of SIRT-1 and resveratrol are still unclear, the studies show that SIRT-1 is essential in lifespan extension and health improvement, and resveratrol promotes SIRT-1 activity.*

Studies in numerous species have demonstrated that reduction of calories 30 to 50 percent below ad libitium levels of a nutritious diet can promote a longer lifespan, improve overall health, promote normal stress resistance and decelerate functional decline. Studies showed that, SIRT-1 activates a critical component of calorie restriction in mammals, and promotes fat mobilization in adipocytes by repressing PPAR (test conducted on mice). Because healthy fat levels have been shown to promote a longer murine lifespan, it is suggested that calorie restriction could be connected to SIRT-1 and life extension in mammals.

As an alternative strategy, new research has focused on the development of calorie restriction mimetics, compounds that mimic the effect of calorie restriction by targeting metabolic and stress response pathways affected by calorie restriction, but without restricting caloric intake. Resveratrol has been suggested as a potential calorie restriction mimetic.  

Supplementing mice with resveratrol significantly increases their aerobic capacity.
